Disease is systemic when immune complexes are formed in the circulation deposited in many tissues e. g. in sle (systemic lupus erythematosus) Disease is localized when immune complexes are deposited in specific tissues e. g. arthritis. Antigen excess persistent infections autoimmune diseases constant inhalation of antigenic material. If we have more antigens than antibodies then fewer fc receptors sticking out of the immune complex available to facilitate removal. Site of deposition (kidney, joints and small blood vessels) possibly favored by high hemodynamic pressures associated with filtration. Instead of being in circulation the complexes are deposited in basement membrane/kidneys. Increased vascular permeability (due to sit of where deposition is) Secrete pro inflammatory substances, enzymes, ros, cause tissue damage. If damage is in circulation this causes platelet activation which forms microthrombi which cause local ischemia as they block the blood vessels.
