Rab5 and rab7: gtp form rab5 overexpressed leads to endosomes that are exceptionally large suggesting rab involved in fusion. Cell expressing mutant rab5 that cant bind gtp lead to small endosomes. Both recruit tethering molecules (eg. eea1 for rab5 ), and attach to membrane, and fuse through synthaxin13. in the case of rab7, recruits hops, and attach to lysosomal or late endosome membrane. Rab5 recruits a complex gef for rab7, which causes rabex5 to stop working. Maturation of early endosomes conversion to mature endosome. Golgi to cell surface: polarized cells have two different domains: basolateral (less cholesterol, depleted in raft components) and apical (rich in cholesterol and sphingolipids, raft material) Basolateral: clathrin vesicles and adaptors proteins ap1 (found on tgn). There are two sub-classes of ap1, one involved in basolateral sorting, while the other involved trafficking of endosomes. Apical: tubular intermediates from tgn, pulled out by motor (kinesin), moved along.
