Bacteria account for most of life"s diversity and exist in almost every conceivable environment, including inhospitable habitats such as the highly saline dead sea. Genetic analysis of bacteria requires special approaches and methods. All prokaryotes are unicellar and lack a membrane-bound nucleus and membrane- bound organelles: eubacteria, archaea. Characteristics of bacteria: diverse shapes and sizes, some are photosynthetic, replication occur prior to binary fission. Bacteria can be grown in liquid medium: grown in test tubes that contain sterile liquid medium, a few bacteria added to a tub, grow and divide until all nutrients are used up. Or until the concentration of their waste products becomes toxic. A growth medium is suspended in gelatin-like agar. Add a dilute solution of bacteria to petri plate. 1 to 2 days, bacteria multiply, forming visible colonies. bacteria: need medium with supplement of other compounds for auxotrophic growth. Complete medium: contain all substances required by all bacteria, including auxotrophic bacteria.
