Cell-mediated immunity: th1 cytokines promote tc killing & macrophage ros killing. In response to il-12 produced by dcs (signal 3) Secrete ifn- (effector cytokine: master transcriptional regulator t-bet, activate cellular immunity, macrophage activation, activation and differentiation of na ve cytotoxic t lymphocytes (ctl) precursors into effector ctl. Important for response to intracellular bacteria: e. g. Important for response to intracellular protozoa: e. g. Signal 3 - ifn- and il-12: these cytokines impact the differentiation of t cells. Ifn- : cytokine produced by differentiated t cells (th1) Tf --> new tf: stat1 & stat4 --> t-bet, new tf --> t cell subtype, t-bet --> th1. Secrete ifn- at point of interaction - at site. Targets macrophages (th1 cell tcr on macrophage surface) Inhibit fusion of phagosome & lysosome: prevent acidification --> lysosomal proteases can"t activate and degrade pathogens, these pathogens are typically those that actively infect and replicate in macrophages.
