Using certain media will encourage growth of certain bacteria over others. Not all bacteria can be stained depends on the cell wall. If stat report is needed specimen is stained before plating. Medically important bacteria will be stained differently depending on the properties of the cell wall (thickness). All bacteria have different cell walls and cell membranes so they are stained differently: gram stain.
