Add mitogens extracellular growth factor; bind to their receptor proteins that are bound on the plasma membrane and trigger signal transduction pathways -> map kinase pathways; triggers phosphylation cascade. Transcription factor can be found in the cytoplasm or nucleus; if in cytoplasm, transports to nucleus. Bind to early response genes: usually transcription factors and stimulate transcription the late response gene. Delayed response gene: 2 cdks, cyclin d and e2f. E2f bind to rb(tumour suppressor: regulating cell dividsion and proliferation) inhibits e2f. As the amount of cyclin e and cdk2 bind up; phosholyate; cause dissociation of rb and e2f. More e2f, more cdk2 and cyclin e, more phosphylation. Remove mitogens from culture solution, remove cyclins; doesn"t matter can pass the restriction point(point where cell can go through synthesis and division) E2f stimulations transcription s phase cyclin cdks, necessary for activation of dna synthesis.
