Physiological effects of eicosanoids (synthesized by body but can also be synthetic) Stim for aggregation release of endoperoxide + txa2 to aggregate platelets. Pgi2 from endothelial cells in blood vessels inhibit aggregation. Labour: incr syn of eicosanoids during labour, cyclooxygenase inhibitors incr gestation + prolong labour but has toxic effect so often use b2 agonist instead. Pge2, pgf2a contract uterus t/out pregnancy and can induce abortions by admin iv, im or intravaginally (preferred so it acts locally) but cannot be used postimplantation (contraceptive) bc would need too large of a dose. Veterinary meds: pgf2a has luteolytic effect (causes regression of corpus luteum) causes period (estrus) in cattle after. Pge2, pgi2 (vasodilators) keep ductus arteriosis in fetus open and indomethacin (cycloox inhibitor) closes it. Ag leads to release of chem mediators like pgs lts. Pge2= bronchodilator pgf2a, tx2, ltc4= response to lts dominates + prolonged bronchoconstriction.
