Human mitochondrial genome: mitochondria have their own genome (mtdna, circular, 16560 bp, encodes 37 genes, heavy (h) and light (l) strand, displacement (d) loop is where replication of it begins. Replication: replicates using binary fission, origin of replication is located in the control region, steps, dna replicates, dna separates, wall invaginates, cell splits, 2 cell now, most genes involved in replication don"t come from the mtdna genome. Contamination: high copy number of mtdna, contamination is huge, sanger uses pcr, pcr amplifies massively, so there"s dna everywhere, pre and post pcr samples can"t be anywhere near each other in the lab. Standard mtdna workflow: contamination so bad the sequences is usually performed on different days, known sample after question sample bc again, confirmation bias. Nomenclature for mutations: reported for their position and mutated base that"s different from refrence.
