Although we discussed a number of reagents in sec ii that react with amino acid side chains, we did not examine specific cases where these reagents were tested against native proteins. In the case of proteins which are denatured (typically with 6m guanidine; or other chaeotropic reagents), we would expect there is little variation between individual residues with the same side chains. However, in the case of the native protein, we would expect that protein microenvironments may play a large role, and could radically alter the reactivity of specific sites. Tyr, lys, or cys sidechains; however, its reaction with these groups is easily reversed by treatment with hydroxylamine allowing determination of the his-modification if appropriate controls are conducted. 50. The authors first determined the pka of individual his residues using 1h nmr.
