Psyc 462 lecture notes 6th feb 2018. Gaba is synthesized from glutamate using enzymatic reaction by glutamate decarboxylase (gad). All 3 gaba reuptake transporters can be found on astrocyte glial cells. Gaba a receptors are ionotropic which allow cl- ions into cell for hyperpolarization. Tiagabine blocks transporter reuptake of gaba. these receptors contain 2 alpha, 2 beta and 1 gamma subunits. Agonist muscimol found in mushrooms which at low doses could cause hallucinogenic effect. Has to work with gaba bound to receptor greater amount of inhibition alcohol binds to same site as benzodiazepine, therefore has sedative effect. Net effect the same as an antagonist. Does not do anything by itself but decreases the efficacy of endogenous ligands can bind to same site as benzodiazepine can produce arousal and seizure. Need 2 similar protein subunits to work side by side. Open k+ channels to have the ions diffuse out of cell along concentration gradient to produce hyperpolarization.
