MICR 302 Lecture Notes - Lecture 7: Lac Operon, Xbp1, Reverse Transcription Polymerase Chain Reaction
Document Summary
Micro 302 lecture 7 - topic 5 (environment) - slides 7-22 continuing with regulatory mechanisms of sigma32. Material for the test will cover topic 1-4 (til the end of proteins) Rpoh mrna increase by 5 fold but transcription of rpoh mrna only increases by 2 fold. 1/2 life increases from about 2 min @ 30 c to 14 min @ 42 c. But: this does not hold true for the mrna induced proteins. Using rtpcr (start with mrna and use reverse transcriptase to make a template, then do quantitative qpcr) Looked at the transcript half-life of dnak and groel. When the proteins were heated up, they were destabilized (not as expected) > they then looked at cspc (cold shock protein) and they found that increasing cspc stabilized the proteins and removing it destabilized them. Specifically: it stabilizes the proteins but had no direct effect of rpoh. Phosphorylation of sigma32: study: (referring to results on slide 9)
