Promoters are not transcribed (at least for 1st year bio there are no exception). The promoter region on the left is upstream of the start point of transcription. Green and black arrows indicate the regions that seem to matter to the. They are the dna bases that come into contact with the polymerase. They tend to cluster around a sq called -10 & -35.
