Document Summary

Required to bind to its promoter and initiate rna synthesis. Their (cid:374)a(cid:373)es i(cid:374)clude (cid:858)tf(cid:859) (cid:894)tfiia, tfiib etc. (cid:895) General transcription factors in binding rna polymerase ii to dna. Tfiid first binds to tata box in the dna sequence. All others primarily interact with each other. Thus, protein-protein interactions play a crucial role in binding stage of eukaryotic transcription. Preinitiation complex a large complex of proteins including rna polymerase ii. General transcription factor tfiih possesses both helicase activity, that unwinds dna, and a protein kinase activity that catalyzes the phosphorylation of rna polymerase ii. Phosphorylation changes the shape of rna polymerase ii, thereby releasing it from the transcription factors, so that it can initiate rna synthesis at the startpoint. Helicase activity unwind the dna, so that rna polymerase molecule can begin to move. Tfiid - ability to recognize and bind to dna promoter sequences is conferred by one of its subunits, tata-binding protein (tbp)
