Chapter 15: dna & gene synthesis and repair. Hershey-chase experiment answers question of whether genes are made up of protein or dna. Dna is made of 1) a backbone and 2) nucleotides that project from backbone. Dna has two antiparallel strands and forms into double helix: strands in 3" (carbon) and 5" (carbon w/ phosphorous) ends. Existing strands of dna serve as template for production of new strands. Deoxyribonucleotides were added to dna strands according to complementary base pairing. & 0. 75 completely new: dispersive replication: stretches of old dna is are interspersed with new dna. Meselson-stahl experiment discovered that dna is semiconservative: they used e. coli since it grows quickly and grew it with 15n. Found that the dna weight was a mixture of 15n & 14n densities (disproving the conservative method: next, the hybrid sample was placed in another medium containing just pure 14n.
