Algal cells- use hemocytometer to count- pipette liquid sample and count cells in the grid. Palmer maloney cells- hold known volume and scan surface of sample to count cells of interest- gives density estimate. Can figure out amount of carbon in sample by burning it ina muffle furnace. Membrane filtration technique- water passed through filter filter places on surface of growth medium incubate count colonies. This is used to detect total coliforms, ecoli coliforms from intestines of warm blooded animals- incubate at 44 degrees. Presence-absence tets- modified mpn test, uses larger sample, sample added to lactose medium add stuff to this- wont tell how many but just tells us that something is present. Defined substrate test- includes idexx system, use mi agar- turns colorless to blue with ecoli- this uses the membrane filter technique. Its important for micribial ecologists to understand what is present and how many are present.
