Pressure
Microbes on land and water surface → 1 atm.
Some bac and arch live in hydrostatic deep sea conditions (barotolerant, barophiles).

Biofilms
Most microbes grow attached to surfaces.
Attached microbes are members of comles, slime-enclosed communities called biofilms

Ubiquitous in nature.
Can be formed on any surface.
Culture Media (Food)
Culture medium is used to grow, transport, and store microorganisms. Agar is an example.
PDA- Potato Dextrose Agar.
Can be solid or liquid.
Must contain all the nutrients required by the organism for growth (major, minor, trace elements, etc).
Types of Media
General purpose media
Support growth of many microorganisms.
Ex: Tryptic soy agar.
Enriched media
General purpose supplemented by blood or other special nutrients.
Ex: Blood agar.
Selective media
Allows growth of specific organisms and inhibits others.
Ex: MacConkey agar tests for Gram negative bacteria.
Differential media
Allows one to distinguish between groups based on characteristics.
Differentiate close relatives.
Ex: Hemolytic versus non-hemolytic bacteria.
Isolation of Pure Cultures
Portobello mushroom exercise.
Groups of cells arising from a single cell.
Allows for the study of a single type of microorganism.
Techniques used to isolate pure cultures: Spread plate, streak plate, and pour plate.
The Growth Curve
Observed when unicellular microorganisms are cultivated in batch culture (liquid).
Has four distinct phases.
