Centrifugation of column based separation based on a property: size, charge, shape of protein etc. First, separate the proteins on gel by change ( rst dimension). (isoelectric. Next, apply rst gel on top of second gel and carry out sds electrophoresis to separate proteins based on size. If one tissue has more of one protein, the spot will appear more of that color. To nd the differential protein expression in two types of cells i. e cancers vs. wild type, label each protein of each cell with either green. If a particular spot is red, cancer cell has more of it etc. Mix proteome of both cells in the same amount. (very similar to microarrays). 2d gel doesn"t tell you what that protein is: identifying and quantifying proteins: Mass spectrometry; an analytical tool used for measuring the molecular mass of peptide fragment. Ionization source: ion source from which charge is transferred to peptide.
