BIOSC 0160 Lecture Notes - Lecture 5: Enzyme, Dna Mismatch Repair, Citrulline
Document Summary
Lecture 5: dna synthesis (cont. : proofreading: dna polymerase iii recognizes mismatched pairs and removed incorrectly paired bases, mismatch repair: newly replicated dna is scanned for mistakes, and mismatches can be fixed, excision repair: the dna is being damaged. It isn"t being replicated it"s sitting and getting damaged in some way, by uvs, etc. Excision repair is a whole system of enzymes that comes in and removes the whole segment that is mutated and dna poly iii or i comes in with ligase and filled in the dna. Chapter 14: working with neurospora because it is eukaryotic and has a nucleus, haploid for its whole life. All of its alleles get expressed as proteins. Diploids don"t express all its alleles, so neurospora being haploid is helpful: there is a specific biochemical pathway to get to arginine. Ornithine and citrulline are needed for arginine synthesis: damaged the dna and tried to find mutants that couldn"t make arginine.
