One of those two strands will be used for the expression of a gene. You can differentiate between the two strands based on the relationship to rna. The newly synthesized rna is complementary to the dna template. One strand is the template strand and the other strand is called the coding strand. The coding strand will have the same sequence as the rna strand except the t will be replaced with a u. These activated precursors will be ribonucleoside triphosphates (utp, gtp, atp, Ctp) which in contrast to dna which needed the diribonucleoside triphosphates. Rna polymerase will also need mg+2 or mn+2 much like dna polymerase. Rna strand will now grow by one residue and pyrophosphate will be released.
