Gtfs are needed for eukaryotic rnap to be able to initiate transcription. Gtfs assemble in the core promoter allowing rnap to get involved and initiate transcription. These are transcriptional factors that are used by rnap 2. They help recognize and initiate transcription at a promoter. There are 6 of these gtfs for rnap 2. How do the gtfs come to recognize the correct sequence and help to get rnap involved? (next slide) It is an emsa assay so it uses a labeled dna piece (this one is a promoter sequence that is strongly expressed from antovirus) and then various proteins are added to that labeled dna. If so, will they retart the mobility of that dna in a gel so instead of seeing the naked dna on the gel, we"ll see bands higher up. There were 5 protein complexes that were tested.
