Diagnosis of parasites: characteristic lesions, presence of parasite & or parasite products (e. g. ova, tests that involve the immune response. Sensitive ability to detect minute amounts of ab or parasite ag. Specific for the parasite (little or no cross-reactivity between different parasites) A wheal (raised, reddened papule or plaque of edema in skin) in immediate hypersensitivity to parasite antigen. Arthus reaction in type iii hypersensitivity (immune complexes) Indurated (very firm -hard) soft tissue lesion seen in delayed type hypersensitivity titer of patient during this step) Incubate with patient"s serum (can do serial dilutions and determine ab. If antibody to parasite ag ispresent, it will cross-link the rbc"s and agglutinate them. Negative result: no agglutination, rbc"s will settle out as small pellet. Incubate with anti-ig (usually anti igg) conjugated to fluorescent dye. Fitc fluorescein isothiocyanate dye will be green under uv scope. Rb200 rhodamine will be red under uv scope. Add antigen and guinea pig complement to each tube.
