When a protein is phosphorylated, they change shape, in which proteasomes can recognize the difference between the phosphorylated protein and the unphosphorylated conformation of protein. Thus, it is possible that proteasomes are targeting the phosphorylated tk2, hence the presence of tk2 in the cell without being phosphorylated.
