Lecture 6 – Cell Surface Receptors and the Tumour Microenvironment
Enzyme-linked Receptors: RTK Family reloaded
• Single Transmembrane proteins
• Many are receptors for growth factors
• Kinase domain on C terminus – induces dimerization & auto phosphorylation
→ provides docking sites for other proteins + drives new reactions
Tumour Microenvironment
• Destroying immune cells that will kill tumours
• Most tumours are heterogeneous – different mutations
• Cell types contributing to the biology of tumours
o Cancer cells – initiate tumours → drive tumour progression
o Cancer stem cells – seed new tumours
o Endothelial cells
o Pericytes
o Immune inflammatory cells
o Cancer associated fibroblasts
o Stromal stem/progenitor cells
• Lysophosphatidic acid (LPA) – activates
downstream growth
Therapeutic Approaches: monoclonal antibodies
(mAbs)
• Also soluble receptors
• Tyrosine kinase inhibitors
• HDAC inhibitors
• E.g. Trastuzumab – HER2/EGFR
• Avastin
• PD1 inhibitors
