Dna sequencing large volume of dna can be sequenced using a dideoxy chain termination method dideoxyribonucleotides are derivatives of the normal deoxyribonucleotide with one crucial difference that is the absence of a oh group on the 3 carbon. Sequencing (dideoxy method) to determine a complete sequence, dsdna is melted and one of the strands is used for sequencing. 4 different ddntps are used in 4 separate dna synthesis reactions.
