Lecture 20"Developmental genetics
Amount of protein binding to enhancer element, is critical to having tight zones of dierentiation.
These end up giving us dierentiation along anterior and posterior areas of the segmentation in
drosophila. !
If we have cells within that segment, we can dierentiation the cells in smaller zones through
mRNA localisation. Individual cell division. localisation of more mRNA in one daughter cell,
compared to the other. Immediately have asymmetric division. !
Induction: A cell has developed a specific function to secrete signalling compound (ligand,
morphogen etc.) and surrounding cells have receptors for the signal compound. Recognition of
stronger signal in adjacent cells to secreting cell. The signal will be less strong, less concentration, !
to cells further away from the secreting cell. !
Over expression experiments: to find if the concentration of BCD (Bicoid) results in the head and
thorax missing. Increase of BCD protein results in further back position of furrow. !
Homeotic gene mutations: mutations that lead to the replacement of one body part by another.
Shows further evidence of the power of transcriptional regulators to influence cellular function.
Determine function by conducting a large scale mutant screen, and looking at the phenotypic
mutation that has resulted from the mutation (e.g. legs where there should be antennae). Isolating
genes which are mutated, and design experiments to ensure that the particular genes are mutated
for the phenotype. Thus, homeobox genes can be separated. !

Document Summary

Amount of protein binding to enhancer element, is critical to having tight zones of di erentiation. These end up giving us di erentiation along anterior and posterior areas of the segmentation in drosophila. If we have cells within that segment, we can di erentiation the cells in smaller zones through mrna localisation. Individual cell division. localisation of more mrna in one daughter cell, compared to the other. Induction: a cell has developed a speci c function to secrete signalling compound (ligand, morphogen etc. ) and surrounding cells have receptors for the signal compound. Recognition of stronger signal in adjacent cells to secreting cell. The signal will be less strong, less concentration, to cells further away from the secreting cell. Over expression experiments: to nd if the concentration of bcd (bicoid) results in the head and thorax missing. Increase of bcd protein results in further back position of furrow.
