BCMB2901 Lecture Notes - Lecture 9: Alternative Splicing, Cleavage And Polyadenylation Specificity Factor, Polyadenylation

36 views2 pages
Lecture 9: Post-transcriptional processing
Processing events:
Capping of 5’ end of RNA (addition of modified G to 5’ end of RNA)
o Cap: a methylated guanine joined to RNA transcript by 5’-5’ linkage
o Linked by 3 phosphates
o 3 sequential enzymatic reactions
o Occurs early
Splicing: cutting & joining segments
o Introns removed, joining exons
Polyadenylation (polyA) of 3’ end of RNA: mediated by poly A polymerase
o CPSF & CstF (cleavage stimualtion factor) bind
o RNA cleavage
o PAP (PolyA polymerase) adds ~200 As at the 3’ end of RNA, using ATP as substrate
Alternative splicing
Same mRNA transcript can be spliced in different ways
è Yields different proteins from same gene
