Bacteria have few membrane bound organelles and grow and divide rapidly; making them great to use for molecular and genetic studies. Plasmids are circular, double stranded dna molecules that usually occur in bacteria. Explain how restriction enzymes work and their importance. A restriction enzyme is an enzyme that cuts dna after recognizing a specific sequence of. The determination of the precise sequence of nucleotides in a sample of dna. 2 methods: sanger method and maxam-gilbert method. Not discovered but an invention by kary mullis. Mutations happen when dna polymerase inserts the wrong nucleotide into a newly synthesized strand. Most common mutation is a point mutation. Snps (single nucleotide polymorphisms) are the most common sources of variation between humans. Most are harmless because they occur in the intron. When they occur in the exons they can affect structure and function. Convert mrnas to cdnas by reverse transcriptase. Each florescent spot represents a gene expressed in the cells.
