Document Summary

Biol 301 laboratory #9: polyacrylamide gels and protein characterisation. Protein sample preparation and loading on 1d sds-page gel. Thaw protein samples on ice: warm the 4x sample buffer at 90-100c x1min before use, use plastic covers next to the heat block to secure microtube lids. Calculate amount of protein sample and ddh2o needed to get 50ul total. Mix 37. 5ul of protein sample with 12. 5ul 4x biffer = 50ul total. Heat samples at 95-100c x5min: 4 tubes for team 1, 5 tubes for team 2, do not heat the mw marker (unstained and coloured, use plastic covers next to the heat block to secure the microtube lids. Centrifuge x15sec at rt: do not put back on ice, keep samples on bench until gel is assembled in the electrophoresis module. Cut open plastic pouch and take gel cassette out. Remove the green tape gently from the bottom of cassette.
