Discard last wash and add 10-12ml of secondary ab soln: be sure to keep the empty vessel. Pour ab soln back into the tube it was provided in (kept in first step) Put the tube in the ice bucket at front of lab: will be stored and reused by other groups. Rinse the membrane with 20ml tbs/t x10sec then discard the tbs/t. Wash membrane 3x with 20ml tbs/t x10min per wash: perform this step on the platform shaker, use fresh tbs/t for each wash, ensure that the membrane is completely covered with tbs/t. Leave membrane in the last wash until you"re ready to perform one of the 2 detection methods: colorimetric, ecl chemiluminescence. Pour off as much of the last wash as possible.
