Bowmans capsule is lined with pec (parietal epithelial cells) Pec do contribute to kidney regen by integration into tissue. Cd133+cd24+ pec cells could divide these cells into pdx+ and pdx- populations. Pdx- = podocalyxin- can diff to tubular duct (and move to duct to contribute to it) and podocyte lineages. Pdx+ = only diff to podocyte lineage, is an intermediate popl toward podocyte lineage. They have an equivalent to metanephric organ, and some nephrogenic zone remaining in adults. Partial nephrectomy can make it respond with de-novo nephrogenesis. Exp: lineage tracing of progen cells in wt and six2-ko cells. So do this with ipsc, step-wise differentiation to kidney organoids ipsc --> primitive streak --> intermediate mesoderm --> metanephric mesenchyme (six2) or ureteric epithelium. Need both metanephric mesenchyme and ureteric epithelium to reciprocally communicate to create kidney organoids!
