Attenuated vaccines since they cannot fight the way the vaccine wants them to fight. Attenuation can be achieved by growing the virus in another organism; then take that newly mutated virus and give back to human to elicit response as a vaccine. If the ag is weak, say dead whole or subunit vaccine, you"d need adjuvant to guide the host in starting response. Slow ag delivery --> more interaction time, more effective. They run around and try to bind the cortical epithelial cells, which express lots of mhci & ii. If the dp t cell can"t bind, they die. If the dp t cell binds just right, it passes. Positive selection and now has to go to single positive depending on what it bound just right to. If bound just right to mhc i, it becomes cd8+
