thMonday January 30 2012 Chagas Disease Biology Immunology and Clinical Aspects Discovery and Distribution Chagas disease caused by T cruzi only found in South America Mainly in Brazil and a little in Peru and Equador as well as the north of Argentina and Chile It is also found in Central America and sometimes in Southern US The difference between this and Leishmania is that the amastigote and promastigote is usually found in Leishmania but here is the trypomastogote and the epimastigote The amastigote and trypomastigote is in the host and the epimastigote form is in the vector Cyclomastigote formis the infectious form So in trypanosoma you have the epimastogote amastigote and trypomastigote form compared to the two in Leishmania To differentiate them the kinetoplasm is where the flagella starts In the trypomastigote you have the kinteoplast black dot on the posterior side of the parasite Where the flagella goes out is the front of the parasite In the epimastigote the nucleus and the kinetoplast are on the same side In the promastigote the nucleus is in the center and the kinetoplast is at the anterior part Then the amastigote form meaning no flagella is round to oval and you have a nucleus and the kinetoplastresidual flagella In the blood Chagas observed trypomastigote forms and amastigote in cells Epidemiology Adobe houses mud blocks mixed with hay are not made very well bugs hide in cracks during the day and come out at night to take bloodmeal You can find 1001000 vectorshouse see slide You will not find vectors over 3000 meters away from where they are Now there are very few people who get sick these days because of efforts to control vectors Keep in mind the vector can hide anywhere not just in poor houses T cruzi infection in human Most people will never know that they were infected until later on sometimes when it is too late They get over it initially and then later on get very sick Trypanosoma cruzi Mophology Form depends on the stage in the blood stage of replication The kinetoplast of T cruzi is so large that it is hard to tell that it is pointed One characteristic of T cruzi is that when you fix it on a slide it has a C shape T brucei will have a more undulating form EXAM HINT FOR THIS SLIDE Undulations in the membrane allow them to swim very quickly in the bloodstream
