*begin by forming gigaseal in the cell-attached con guration as discussed in last lecture. What do we mean when we say: seal is both mechanically and electrically tight? : the ions are only owing though the channel because they cannot pass through any other part due to mechanical seal created by suction. Most basic con guration is on-cell or cell-attached patch- forms of basis of all other con gurations- electrode is sealed to patch of membrane and rest of cell is intact- Other con gurations : whole-cell clamp (including perforated-patch variation, inside-out patch, outside-out patch. Note: advantages and disadvantages of each con guration- each con guration is appropriate for the type of channel (and its regulation) under study. Procedures for obtaining remaining three con gurations (from cell-attached patch) : whole-cell- strong suction- pipette has direct access to the cytoplasm, inside-out .
