Immuno Lecture 3/4 Notes- September 25/ 2012 Lecture 3- Innate Immunity I  Note: Sterile infection= when the bodys own cells are all dying for example Recognizing Danger  PAMPs= Pathogen Associated Molecular Patterns (ligand) o Not on host cells (therefore microbial pathogens) o Small number of receptors for a large number of pathogens o PAMPS= KEY TO THEIR SURVIVAL o Common/invariant structures  DAMPS= Danger Associated Molecule o HMGB- high mobility box group= stabilizes chromatin o HSP= heat shock protein o ATP, uric acid o Injury, cell death , stress  Receptors: Phagocytic Receptors (cause phagocytosis) and Pattern recognition Receptors o Phagocytic= no signaling= mannose receptor, scavenger receptor, complement receptor o Pattern Receptors= NLR (NOD-like receptor), NALP, Toll-like receptor, Mannose Binding Lectin Signaling Pathway  TLR is activated by PAMPS/DAMPS  MyD88= activated= induces transcription of inflammatory cytokines (TNFα, IL-6, IL-1β  TRIF (more limited to TLR3 and TLR4)= increases transcription of type 1 interferon (IFNα and IFNβ) Localization of TLRs (Innate receptors)  TLR2/TLR6 and TLR1/TLR2 = called TLR2 mediated signaling = on cell surface  Note: TLR2 cannot signal alone= once activated= dimerizes with either TLR6 or TLR1  TLR5, TLR 4 and TLR 11 also found on the membrane  Inside in an endosome= TLR3 (dsRNA), TLR7(ssRNA) and TLR9 (CpG DNA) Note: Important features about the innate immune system:  Innate receptors already present= an immediate response  Without the adaptive immunity you can still fight off many infections  Ie. Without the innate immunity = you will die NK Ce
