BCHM 316 Lecture Notes - Lecture 17: Phosphopantetheine, Transferase, Acetyl Group

56 views2 pages
Before the condensation reactions that build up the fatty acid
chains can begin:
TWO thiol groups that are on the enzyme complex must be
charged with the correct acyl groups!
So like sH on Ks and sH on ACP
PRIMING STEPS
1. The acetyl group of acetyl-CoA is transferred to the ACP in
a reaction catalyzed by the malonyl/ acetyl-CoA-ACP
transferase (MAT). Afterwards, the acetyl group is then
transferred to the S-H group of β-ketoacyl-ACP synthase
(KS)
2. Malonyl group transferred from malonyl-coA to the SH
group of ACP; this is catalyzed by malonyl/ acetyl-CoA-
ACP transferase (MAT)
FA SYNTHESIS STEPS
1. CONDENSATION
2. REDUCTION
3. DEHYDRATION
4. REDUCTION
Condensation: condensation of activated acetyl and malonyl
groups to form acetoacetyl-ACP; co2 produced; catalyzed by β-
ketoacyl-ACP synthase
Acetoacetyl-ACP : an acetoacetyl group bound to ACP through the phosphopantetheine SH group
The acetyl group is transferred from the Cys-SH group of the
enzyme to the malonyl group on the SH of ACP

Document Summary

Before the condensation reactions that build up the fatty acid chains can begin: Two thiol groups that are on the enzyme complex must be charged with the correct acyl groups! So like sh on ks and sh on acp. Priming steps: the acetyl group of acetyl-coa is transferred to the acp in a reaction catalyzed by the malonyl/ acetyl-coa-acp transferase (mat). Afterwards, the acetyl group is then transferred to the s-h group of -ketoacyl-acp synthase (ks: malonyl group transferred from malonyl-coa to the sh group of acp; this is catalyzed by malonyl/ acetyl-coa- Fa synthesis steps: condensation, reduction, dehydration, reduction. Condensation: condensation of activated acetyl and malonyl groups to form acetoacetyl-acp; co2 produced; catalyzed by - ketoacyl-acp synthase. Acetoacetyl-acp : an acetoacetyl group bound to acp through the phosphopantetheine sh group. The acetyl group is transferred from the cys-sh group of the enzyme to the malonyl group on the sh of acp.
