Factors include: gut microbiota, age & sex, diet & environmental factors, diseases/physiological states, individual differences & drug-drug interactions. As well as: species differences, hereditary, induction/inhibiton, drug dose. So rst off, note that the gut microbiota needs to be better studied. These may result in a different metabolite pro le than intended due to gut microbiota , we have unexpected activation/ deactivation. So as you get older, there is less cyp activity. As well, when you get older there are changes in liver size and perfusion of the liver. Females have differences in hepatic expression in many enzymes like cyp 3a4 and less gut enzymatic activity. There are also diet and environmental factors some food/environmental factors affect the expression/ action of these enzymes. There are also phase 2 cofactors that are dependent on diet. Liver cirrhosis, hepatitis, liver cancer same for other organs as well: pregnancy. Great changes in expression of various drug metabolizing enzymes: placental expression as well.
