BCH 361 Lecture Notes - Lecture 10: Heat Shock Protein, Peptide, Pancreatic Ribonuclease
Document Summary
The protein data bank (pdb) and visualization software. The pdb contains all experimentally determined structures of macromolecules. Each structure given an id number: pdb id. Motifs can be simple or complex: a protein domain is part of a polypeptide chain that is independently stable, has a particular, independent function from the rest of the polypeptide, and often appears in a variety of proteins. Domains can be comprised of a single motif or several simple motifs. Since each type of domain has a particular function, one can mix and match domains to couple functions. It is mathematically impossible for protein folding to occur by randomly trying every conformation until the lo(cid:449)est energ(cid:455) one is found (cid:894)le(cid:448)inthal"s parado(cid:454)(cid:895) Search for the minimum is not random because the direction toward the native structure is thermodynamically most favorable. Energy landscape of protein folding -collapsed structures give rise to molten globules partially formed domains and motifs.
