MBB 231 Lecture Notes - Lecture 12: Protein Disulfide-Isomerase, Differential Centrifugation, Disulfide
Document Summary
Endomembrane system: know what organelles make up the endomembrane system. Rough er, smooth er, golgi complex, endosomes, lysosomes: understand diferenial centrifugaion and density-gradient (rate-zonal) centrifugaion and be able to apply these techniques theoreically. Separaion of cell organelles by size/density by diferent rates of sedimentaion. Technique for separaing organelles based on diferences on sedimentaion rate. The sample for fracionaion is placed as a thin layer on top of a gradient of solute that increases in density from top to botom. Organelles move into the gradient unil each reaches equilibrium (buoyant density) , it"s the point where the density of each organelle is exactly equal to sucrose density: know four aciviies the smooth er paricipates in. Involved in: drug detoxiicaion, carbohydrate metabolism, calcium storage, steroid biosynthesis. Involved in the formaion of transiion vesicles that shutle lipids and proteins from er to golgi. Step 1: acyl transferaces successively add to faty acids to glycerol phosphate to produce phosphaidic acid.
