The human body is designed to be resistant to harm by foreign invaders and toxic substances but unfortunately these same characteristics may limit the ability of drugs to achieve clinical effectiveness. In order for a drug to be effective it must be able to cross the physiologic barriers that limit the access of foreign substances to the body. Drug absorption must first occur by mechanisms that are designed to bypass these barriers. Following absorption, the drug needs to be distributed throughout the body via systems such as the blood or lymphatic vessels to reach its target organ. The last two processes will affect the drug"s ability to access its target within the body. Metabolism involves inactivation of the drug through enzymatic degradation (liver) while excretion involves elimination of the drug from the body (kidneys, liver and feces). In this unit we will provide an overview of these pharmacokinetic processes.
