Ab-based methods: western blotting, microscopy methods, cell counting/cell staining. Immunoelectron microscopy: to determine the location in/on a cell, without damaging the cell or losing components, ect. Now the protein has these two epitopes on the end. Take images under excitation wavelength for each individual tag. Fluorescence-based cell counting and cell sorting: mark specific cell types with different fluorescent tags + use those tags for either counting or sorting. Flow cytometry: counting cells with particular fluorescent patterns, 8 to 16 different colours that you can look at, at the same time. Go to the computer, collect data and measure out of all the tcr+ cells which ones have cd25+ ect. Basically flow cytometry with collecting the cells at the end.
