Lymphocytes, present in blood, are for the most part small cells (6 in diameter) with little cytoplasm and are not normally dividing. When induced to divide by mitogens, they first increase in size (up to a diameter of 15 ) with a concomitant increase in their ribosomes, endoplasmic reticulum and mitochondria, see figure 1. A major route is via the thoracic duct (see below). Gowans showed in the early 1960"s that animals can be depleted of lymphocytes by draining them from the thoracic duct, whereupon they become immune-deficient (gowans, about 1960). Reconstitution with lymphocytes from syngeneic donors restores immunocompetence, and reconstitution with lymphocytes from an animal primed to a particular antigen allows the reconstituted animal to mount a secondary response on challenge with this antigen.
