CHEM237L Lecture Notes - Chromatography, Histidine, Lysine
Document Summary
Electrophoresis: the actual copy of the electrophoregram is with my partner amreen. I have included a sketch version of what it looked like on my report form for week 2. Lysine was expected to elute from the cation exchange resin in the basic zone (last) because it has a strong affinity for the ions inside the column. At a ph of 4. 2 its net charge was +1 and at a ph of 13 its net charge was -1, confirming that it will take a longer time to be eluted than the additional 4. Lysine was expected to travel the farthest during the paper electrophoresis because at ph 2 its net charge is +1. 6 and the amino acids are travelling from anode to cathode; it had the largest positive charge. When lysine is mixed with dtnb the expected result is expected to be negative (remain the same colour prior to exposure) because it doesn"t have any sulfhydryl group.
