Eliminaion: excreion of unchanged drug removal and eliminaion of parent drug. Therefore, metabolism clears or eliminates the parent drug, even though metabolites may sill be in the body. Uptake into the issues is never clearance no mater how slowly the drug comes back into systemic circulaion. Deiniion of clearance: volume of blood cleared of drug per unit ime (ml/min or l/hr) Extracion raion: the value of 0. 5 for the drug (how much is cleared in one pass of the liver) Extracion raio (eh) = 1 (concentraion of drug leaving liver/concentraion of drug entering liver) Clearance depends on blood low to the eliminaing organ (qh) and eiciency of extracion of that organ (eh) (note: eh = hepaic raio) Another deiniion of clearance: the constant relaing drug concentraion in plasma to the rate of eliminaion in the body.
