A role for micrornas in the drosophila circadian clock. A series of experiment was made to understand the contribution of translational control by microrna to circadian rhythm in drosophila. First, the mirna biogenesis pathway was knocked down in drosophila circadian tissues. Specifically, an inverted repeat (ir) transgene of dicer-1 was combined with tim-gal4 and uas-cycvp16, a circadian-mediated transcription. The presence of dicer-1 ir inhibited the mirna biogenesis resulting in the disruption of drosophila behavioral rhythms. This indicated that mirna biogenesis was necessary for circadian rhythm. Next, an in vivo method was developed to identify the mirna-mrna pairs involved in the regulation of circadian rhythm. Immunoprecipitation (ip) of ago1, a protein involved in the. Risc complex, followed by microarray analysis of the extracted mrnas from the ago1-ip identified several mrnas under mirna control, some of which were predicted to have more than one mirna-binding sites by the pictar algorithm.
