Document Summary

Preparation of proteins for sodium dodecyl sulfate-polyacrylamide gel electrophoresis (sds-page) Week 9: protein expression in wild type and lac operon mutant strains of e. coli using. There are several different biochemical techniques that can be used to purify and characterize the macromolecules of a cell. For the purposes of this exercise we will concentrate on proteins. Proteins have different sizes, composition, solubility in aqueous solutions as well as other unique physical or chemical properties that can be used to differentiate protein populations. In order to study the characteristics or specific proteins in a cell (if unicellular) or tissue (multicellular), you must collect the material of interest and lyse the cells to release the proteins. In addition, you may wish to perform subcellular fractionation of eukaryotic cells to allow isolation of particular organelles (such as the nucleus or mitochondria) to assay proteins unique to a particular organelle.
