BIOC14Lec 10Article 2 Discussion only sections 1 2 Article 2 Genetic Dissection of Neural Circuits we are looking at the relationship btn genes and behaviors in terms of intervening things ex neuron itself as the unit of activity in the brain transgenic and knockout mice for generating mice which lack certain genes or certain genes addedbased on these models we can assess the effect of gene problem when you knockout a gene embryonically can have all kinds of compensatory effects and other effects if gene is required for normal development and you knock it out wont be able to test them behaviorally in adulthood because they wont survivetherefore you want some control over when this gene is knocked outexpressed diff methods for generating mice this kind of technology used in other model organism like Drosophila fruit flies but mouseclassic model DNA microinjection into singlecell embryos as a way to produce transgenic mice with multiple copies of randomly inserted DNA VS specifically targeting certain regions of the genome via homologous recombination to generate replacements at very specific loci studying individual genes isnt sufficient for studying how the brain works and studying individual cells is also not sufficient bc cells work in groupscircuitsidentify circuits by class neurons into types depending on what theyre doing until 1950s we had to stain cells and look at its morphology but now we can identify how the neurons perform in circuits their electrical activity identify specific patterns of gene expression in particular cell types and distinguish them from others and can identify patterns of connectivitywhere they synapse w other neurons we want 2 be able to modulate their activity specifically one of the fruitful ways of identifying subtypes of genes is by their expression patternsusing RNA in situ hybridizationway of preserving mRNA in tissues and labeling with a probe that will hybridize to specific sequences and probe can be labeled w radioactivityfluorescent dyeantigen that you can then label w something else
