The navarre lab has stocks of many different ingredients to make many different types of food for microbes (shows pic of a variety of dried media ingredients). To make bacterial growth media we first add the basic nutrients in the required amounts and dissolve in water. It is like gelatin but has a higher melting temperature (shows clip of adding 25g of nutrient powder and then 7. 5g of agar and then mixed together). Pure water is added to a final volume of 500 ml to dissolve the ingredients (agar does not dissolve unless it is heated). A bar magnet is placed inside the flash to stir the ingredients when it is placed on a heated stir plate . The magnetic bar will be left in the flask during sterilization and will help when we mix in other ingredients. The media is sterilized in an autoclave at 121oc for one hour. This will both kill contaminants and dissolve the agar.
