Potency: really just means how little drug is required to have the desired effect, more/better binding interactions between evolving drug and target = improving potency, better match with target = good potency. Once we understand a molecule and how/why it works, then we can start making it. Let"s say we isolated this compound from the nasal secretions of an unusually hairy. Hebridean bogwoppit and think it might be a treatment for baldness. The compound cannot be synthesized economicallyand it absolutely stinks: however, this species of bogwoppit is exceptionally vicious and cannot be domesticated. Easiest thing - take the staring compound and remove or modify each group - then see if it make a difference to the activity loss of activity = essential no change in activity = part of structure not necessary. Structural extension to increase potency and nd additional binding interactions.
