Pten: when knocked out, you get accelerated cell growth, it is very well-characterized and detailed critical pathway toward tumour genesis and cancer cell survival and progression. Loss of function mutations in cancer means that it cannot be the target because it is already inactivated. If its activity is regulating the known pathway, you have to figure it out and those become the targets. Once they realized that this is a critical molecule that is affecting tumour-genesis, they had to go in and look at the biochemistry and cell biology of the enzyme. Over expression systems were fundamental to this. Bioinformatics: looks like it is a tyr phosphatase enzyme. When testing it, they used an in vitro expression system. Something about the results that told the researchers that it is probably not the target.
