Topic 23We got primary transcript made before that can be translated that primary transcript must be porcessedYou dont make pirmary transcript having sitting around and gets 5 caped and 3 pol adenylated all processes happen as soon as they can As soon as 5 end is made and comes out of rna polymerase it almost right away gets capped All processes are concerted with each other and with transcriptionRna transcript doesnt sit around and wait to be capped and adenylatedPrimary transcripts becomes a mRNA for export out of the nucleus and translation 5 capping 3 polyadenlation Splicing Concerted processeshappen togetherRequire specific enzymes for these processesPrimary transcript becomes a mRNA for export out of the nucleus and translation 3 processing events These processes are concerted so they are all happening at the same time as transcription We got pimary transcriptdont memorize strucutrebut there s 7methyl quanosine cap on transcript Here is our primary transcript and we got 5 end To that we add 55linkagethat is a 55 linkage of 7 methyl guanosine Thats the cap Why bother with this process Marks the 5 end of the mRNA as being intact As soon as it comes out of polymerase it is capped and then all machinery knows that is an authentic 5 end The 5 end is required for mrna epxport and translation You only want to translate and transport intact rna requried for mRNA export and for translation We got 5 cap transcript and stop codonDownstream sequence aauaaathis recruits complex that does a few things Clips rna at that point and other enzymes in the complex that just down stream long poly a tailWe got 5 end that is capped We got aauaaathis recrutis complex Complex clips rna at that point and therer are enzymes in the complex that add poly a tailthe tail can be around 300 bases in length There are enzyme that recognize the aauaaa sequnececlips downstream and poly a tail is added what does the poly a tail do helps protect the 3 end of mrna from degradation in cell there are 3 endonucleases that chew rna from 3 endbut having long poly a tail at end helpes protect rna from degradation
